For those that play the NL cash tables, I think the big overbet is almost always a sign of someone on a complete bluff or with a weak hand like middle pair or top pair on a dangerous board. So long as I've got a pretty good read on the player I'll call that (provided I have a good hand) under the right circumstances (wrote more about this in my blog a couple weeks back).
